TARGINACT® (Oxycodone Hydrochloride and Naloxone Hydrochloride Prolonged Release Tablets) is a prescription opioid combination analgesic indicated for the management of severe persistent pain in adults requiring opioid analgesics for adequate pain control. The combination of oxycodone and naloxone provides strong analgesia while reducing opioid-induced constipation (OIC) through peripheral intestinal opioid receptor antagonism.
- Mechanism: Oxycodone activates central μ-opioid receptors to provide potent analgesic effects. Naloxone selectively blocks peripheral intestinal μ-opioid receptors to reduce opioid-related constipation without significantly affecting central analgesia.
- Drug class: Opioid analgesic combination (controlled narcotic medication).
- Clinical use: Long-term management of severe cancer pain and other severe chronic pain conditions requiring opioid therapy.

How to use ?
Recommended Dose: The recommended starting dose is 5 mg/2.5 mg every 12 hours, followed by gradual dose adjustment according to pain severity and individual tolerance.
Time: Administer every 12 hours at fixed times, usually morning and evening.
Instructions for use: Swallow the tablet whole with water. Do not crush, chew, split, or grind the tablet, as damaging the prolonged-release structure may cause rapid release of oxycodone and increase the risk of severe respiratory depression. Treatment should be individually titrated and discontinued gradually after long-term use to avoid withdrawal symptoms.
Interaction: Avoid concurrent use with alcohol, benzodiazepines, sedatives, hypnotics, other opioids, or monoamine oxidase inhibitors (MAOIs) due to increased risk of respiratory and central nervous system depression.
Side effects: Common adverse reactions include drowsiness, dizziness, nausea, vomiting, dry mouth, fatigue, constipation, sweating, and hypotension. Serious reactions may include respiratory depression, dependence, and withdrawal syndrome.
