 Indication & Scope

TRUQAP® Capivasertib Tablets combines use with fulvestrant for adult patients with ER-positive, HER2-negative locally advanced or metastatic breast cancer carrying PIK3CA/AKT1/PTEN gene alterations that progress after prior endocrine therapy.

  • Mechanism: Reversible selective pan-AKT kinase inhibitor that blocks AKT downstream oncogenic signaling pathway, inhibits tumor cell proliferation induced by PI3K/AKT pathway mutations and reverses endocrine therapy resistance.
  • Drug class: Oral targeted AKT inhibitor, anti-tumor prescription small-molecule medicine.
  • Clinical use: Second-line combination targeted therapy for endocrine-resistant mutated advanced breast cancer.

How to use ?

Recommended Dose: 400mg twice daily for 4 consecutive days weekly, followed by a 3-day drug-free break throughout each treatment cycle.

 

Time: Doses are taken 12 hours apart at fixed daily times, combined with scheduled fulvestrant injections.

 

Instructions for use: Swallow tablets whole without crushing or chewing; can be taken with or without meals. Dose reduction is allowed for grade 2 and above adverse metabolic reactions.

 

Interaction: Strong CYP3A4 inhibitors increase systemic exposure of capivasertib, while strong CYP3A4 inducers reduce therapeutic efficacy, concomitant use should be avoided.

 

Side effects: Diarrhea, hyperglycemia, fatigue, nausea, stomatitis, rash, elevated liver enzymes, and decreased appetite.
