 Indication & Scope

Klacid® is a prescription macrolide antibiotic indicated for the treatment of infections caused by clarithromycin-susceptible microorganisms, including upper and lower respiratory tract infections, skin and soft tissue infections, Helicobacter pylori eradication therapy, and atypical bacterial infections.

  • Mechanism: Clarithromycin binds to the bacterial 50S ribosomal subunit and inhibits protein synthesis, suppressing bacterial growth.
  • Drug class: Second-generation macrolide antibacterial agent.
  • Clinical use: Treatment of susceptible respiratory infections, atypical infections, and combination therapy for H. pylori eradication.

How to use ?

Recommended Dose: Adults: Common infections: 250 mg every 12 hours. Severe infections: 500 mg every 12 hours. Usual treatment duration: 7–14 days. For Helicobacter pylori eradication: 500 mg twice daily combined with proton pump inhibitor and another antibacterial agent for 10–14 days.

 

Time: Take orally with or without food. Swallow tablets whole with water.

 

Instructions for use: Complete the prescribed course. Dose adjustment is required in severe renal impairment. Do not use alone for H. pylori eradication because monotherapy rapidly induces resistance.

 

Interaction: Clarithromycin strongly inhibits CYP3A4. Avoid combination with simvastatin, lovastatin, ergot derivatives, cisapride, pimozide, and astemizole. Monitor patients receiving warfarin, digoxin, or other CYP3A4 substrates.

 

Side effects: Common adverse reactions include nausea, diarrhea, abdominal discomfort, vomiting, taste disturbance, headache, dizziness, rash, and elevated liver enzymes. Serious adverse reactions may include severe allergic reactions, QT prolongation, liver injury, Clostridioides difficile–associated diarrhea, severe skin reactions, and blood abnormalities.
