Indication & Scope
AnXi® (Rovadicitinib) is a Class 1 innovative small-molecule kinase inhibitor developed by Chia Tai Tianqing Pharmaceutical Group for the first-line treatment of adult patients with intermediate-2 or high-risk primary myelofibrosis (PMF), post-polycythemia vera myelofibrosis (PPV-MF), or post-essential thrombocythemia myelofibrosis (PET-MF) Rovadicitinib targets JAK and ROCK signaling pathways to reduce disease-related splenomegaly and improve myelofibrosis-associated symptoms.
- Mechanism: Rovadicitinib selectively inhibits JAK1/2 and ROCK1/2 kinases. By blocking JAK-STAT signaling, it reduces inflammatory cytokine activity; inhibition of the ROCK-STAT3 pathway may reduce fibroblast activation associated with myelofibrosis.

How to use ?
Recommended Dose: 5 mg administered orally once daily according to the approved prescribing information.
Time: Administer once daily on an empty stomach.
Instructions for use: Swallow the tablet whole. Dose adjustments may be required based on blood cell counts and treatment tolerance. For patients experiencing severe thrombocytopenia or anemia, dose reduction to 5 mg every other day may be considered according to clinical evaluation.
Interaction: Strong CYP3A4 inhibitors may increase rovadicitinib exposure and require dose adjustment. Strong CYP3A4 inducers may reduce therapeutic exposure and should be avoided.
Side effects: Common adverse reactions include thrombocytopenia, anemia, neutropenia, nausea, headache, upper respiratory tract infection, and increased blood lipid levels.
