Indication & Scope
Hernexeos® Zongertinib Tablets treats adult patients with unresectable or metastatic non-squamous non-small cell lung cancer harboring activating HER2 tyrosine kinase domain mutations detected by authorized diagnostic tests.
- Mechanism: Zongertinib is an irreversible selective HER2 tyrosine kinase inhibitor. It covalently binds to cysteine residue C805 within the HER2 kinase domain, selectively blocks HER2-driven proliferative signaling with minimal off-target EGFR inhibition, suppressing HER2-mutant lung cancer cell proliferation and metastasis.
- Drug class: Selective HER2 tyrosine kinase inhibitor; oral targeted anti-cancer small molecule.
- Clinical use: First-line and post-platinum chemotherapy targeted treatment for HER2-mutant advanced NSCLC; demonstrates robust anti-tumor activity for patients previously treated with HER2 antibody-drug conjugates.

How to use ?
Recommended Dose: 60 mg taken orally once daily for adult patients.
Time: Consistent daily dosing at the same hour, can be administered with or without food.
Instructions for use: Swallow tablets intact with plain water; do not split or crush. Monitor pulmonary function and liver enzymes monthly throughout treatment.
Interaction: Strong CYP3A4 inhibitors significantly elevate zongertinib plasma concentration, requiring dose reduction; proton pump inhibitors do not alter overall drug absorption efficacy.
Side effects:
1. Common reactions: diarrhea, fatigue, stomatitis, paronychia, dry skin, elevated transaminases.
2. Severe rare adverse events: interstitial lung disease, grade 3 diarrhea, severe cutaneous toxicity.
