 Indication & Scope

Glucotrol XL® is a prescription second-generation sulfonylurea indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus when lifestyle interventions alone are insufficient. It can be used as monotherapy or in combination with other antidiabetic agents, including metformin, insulin, and DPP-4 inhibitors. The osmotic pump controlled-release formulation provides gradual drug release over 24 hours, allowing once-daily dosing.

  • Mechanism: Glipizide stimulates insulin secretion from pancreatic β-cells by closing ATP-sensitive potassium channels, increasing insulin release and lowering blood glucose.

How to use ?

Recommended Dose: The usual starting dose is 5 mg once daily. The dose may be gradually increased according to blood glucose response. The maximum recommended daily dose is 20 mg.

 

Time: Take once daily before breakfast.

 

Instructions for use: Swallow the tablet whole. Do not crush, chew, or split. The empty tablet shell may appear in stools and is a normal result of the controlled-release system.

 

Interaction: The risk of hypoglycemia may increase when combined with insulin or other glucose-lowering agents. Corticosteroids, thyroid hormones, and some diuretics may reduce the glucose-lowering effect.

 

Side effects: Common adverse reactions include hypoglycemia, nausea, abdominal discomfort, dizziness, headache, and skin reactions.
